Assessment of DNA Damage of Oral Mucosa due to Ceramic Bracket Using Comet Assay and Mutagenicity of Orthodontic Bonding System Using Ames Test

Objective: The objective of this study was to determine the DNA damage caused by ceramic bracket (Clear II, SIA Orthodontic Manufacturer Srl, Italy) on human buccal mucosal cells using comet assay and mutagenicity orthodontic bonding system (Transbond XT; 3M Unitek) Ames test. Methods: In study, twenty patients were recruited from Specialist Clinic, Hospital Universiti Sains Malaysia. Buccal cell sample obtained each patient at three time points-before (TO), after one month (T1) two months (T2) placement for performing assay. spot test version performed four Salmonella Typhimurium (S. Typhimurium) tester strains (TA 98, TA100, TA1535 TA1537) testing. Total score (TCS) frequency (DF) used non-parametric Friedman followed multiple pairwise comparison. analysed a non-statistical method based revertant growth ring formation. Results: There no significant change both TCS DF between T0 T1, but parameters increased significantly T1 T2. Non-statistical analysis carried out evaluate results formation colony ring. None S. showed any around light cure adhesive primer. Conclusion: Ceramic does not cause primer is non mutagenic under present conditions.
